Sequencing
The Core currently processes samples primarily on the
ABI Prism 3700 Automated DNA Analyzer for both Sequencing and Genotyping
applications and also owns a Model 377 which is sometimes used for backup
capacity. The 3700 is designed for high throughput runs and is capable
of processing up to four 96-well plates a day as well as handling 384-well
plate formats. The ABI 3700 Sequence Analyzer is a fully automated system
that allows for the unattended analysis of multiple 96-lane runs. Samples
are electrophoresed through a system of capillaries using a polymer
matrix.
Sequencing runs are done on Tuesdays and Thursdays.
Average turn around time is 2-4 days depending on the workload. 500-800
base length reads are expected from one sequencing reaction, these results
may vary depending on the particular template. We require your DNA samples
to be submitted in 200 ul PCR tubes. Total volume should always be 6
ul either for plasmid or PCR template. We need 50 ng of DNA per 1kb
of your product and 2 pmol of the primer. The ABI 3700 Genetic Analyzer
used for sequencing is highly sensitive so it is crucial that DNA samples
are purified and free from residual salts, proteins, residual detergents,
and RNA. All samples must be in water - NO buffers. We need duplicates
of your samples, so that we have the possibility to repeat the sequencing
reaction if we need to. We provide the following standard primers at
no cost. Please check the sequence with those in your sample to ensure
they are the same. Some companies have made changes without changing
the name of the primer.
